Modified Differential Medium for Identification of Salmonella and Shigella

Abstract
|

|
To increase the detection rate of Salmonella and Shigella spp. from carrier and contaminated environmental material, we make anew differential medium suitable to growth and isolation of Salmonella and Shigella spp.
This new differential medium was made by adding the yeast extract and beef extract to MacConkey agar medium and was the colony productivity compared between the new medium and conventional MacConkey and S-S agar medium. We add oxgall in various
concentration to BHI agar medium and investigate the effect of the oxgall concentration on the growth of Salmonella and Shigella spp. the following results are obtained
1. Salmonella and Shigella spp. grow well on MacConkey-yeast extract-beef extract agar medium and form a large colonies.
2. MacConkey-yeast extract-beef extract agar medium is better than MacConkey or S-S agar medium as a differential medium of Salmonella and Shigella spp.
3. The colonies of Salmonella typhi were larger in a medium with oxgall than those in a medium without oxgall.
